What is a Semen Analysis Test?

A semen analysis, also called a sperm count test, measures the quantity and quality of semen and sperm. Semen is the thick, white fluid released from the penis during a sexual climax called ejaculation. Semen contains sperms, made in the male reproductive system that include the genetic material needed to conceive.

Why is the Semen Analysis prescribed?

The semen analysis is the single most important test for investigation in cases of male infertility. It is recommended to find out the cause of infertility in case a couple is facing difficulty in conceiving. A problem with semen in quality or quantity may be one of the major reasons for infertility. Problems such as a low sperm count or compromised immobility of sperm can cause infertility.

This test is also done after VASECTOMY (a procedure done in males for permanent sterilization) to ensure that sperm are not present in semen. Abnormal test results could indicate multiple problems like hormonal imbalance, infection, or radiation exposure.

Precautions/prerequisites for Semen Analysis:

For Semen analysis, the patient should practice Sexual Abstinence or Refrain from Ejaculation for 2-5 Days (Ideally 3 Days) before Sample collection. A specimen can be obtained by Masturbation in a wide-mouthed, sterile, dry plastic container.

For best results, the patient should urinate, wash & dry Genital organs, and collect the entire Ejaculate. Avoid alcohol, Caffeine, Smoking, and other medications before the test.

Do I need to go to the lab for Semen Analysis? 

You need to visit the lab to give a Sample for Semen Analysis. In case you have collected a sample at home, it should reach the lab within 15 to 20 minutes after collection.

The sample should be kept at body temperature before being it to the lab for examination. You may need to provide more than one sample within a week or two. That’s because your sperm count and semen quality can vary from day to day.

Are there any specific instructions for transporting the semen sample to the lab?

Transporting a semen sample to the lab requires specific steps to ensure the integrity and accuracy of the sample. Here are the key instructions:


  1. Timing: Ideally, the sample should be collected and delivered to the lab within 30 to 60 minutes.
  2. Container: Use a sterile, wide-mouthed, plastic container provided by the lab. Do not use condoms or other containers, as they can contain spermicides or other chemicals that may affect the sample.
  3. Abstinence: Maintain 2 to 7 days of sexual abstinence before collecting the sample to ensure optimal sperm quality.

Collection Method:

  1. Hygiene: Wash your hands and genital area thoroughly before collecting the sample.
  2. Method: The sample should be collected by masturbation. Avoid using lubricants, as they can contaminate the sample.
  3. Complete Sample: Ensure the entire ejaculate is collected in the container, as the first part of the ejaculate contains the highest concentration of sperm.


  1. Temperature: Keep the sample at body temperature (around 37°C or 98.6°F). You can transport it close to your body, such as in a pocket or under your arm.
  2. Avoid Extreme Temperatures: Do not expose the sample to extreme temperatures (hot or cold), as this can affect sperm motility and viability.
  3. Time Sensitivity: Deliver the sample to the lab as quickly as possible, ideally within 30 to 60 minutes after collection.

  1. Label: Ensure the container is properly labeled with your name, date of birth, and the time of collection.
  2. Form: Fill out any required forms or documentation provided by the lab.
